The only door the Losties opened was the Hatch so the statement above indicates they know about it.

Or he could have just been making a general analogy about sticking their nose where it doesn't belong.

But I think your right. I think he was talking about the Swan. But this still doesn't mean that they knew of the Swan's location or what was going on inside it before Locke and Boone found it.
